Dense-core secretory granule biogenesis.
The dense-core secretory granule is a key organelle for secretion of hormones and neuropeptides in endocrine cells and neurons, in response to stimulation. Cholesterol and granins are critical for the assembly of these organelles at the trans-Golgi network, and their biogenesis is regulated quantitatively by posttranscriptional and posttranslational mechanisms.
متن کاملLarge dense-core secretory granule biogenesis is under the control of chromogranin A in neuroendocrine cells.
The large dense-core secretory granule is an organelle in neuroendocrine/endocrine cells, where prohormones and proneuropeptides are stored, processed, and secreted in a regulated manner. Here we present evidence that chromogranin A (CgA), one of the most abundant acidic glycoproteins ubiquitously present in neuroendocrine/endocrine cells, regulates dense-core secretory granule biogenesis. Spec...
متن کاملChromogranin A, an “On/Off” Switch Controlling Dense-Core Secretory Granule Biogenesis
We present evidence that regulation of dense-core secretory granule biogenesis and hormone secretion in endocrine cells is dependent on chromogranin A (CGA). Downregulation of CGA expression in a neuroendocrine cell line, PC12, by antisense RNAs led to profound loss of dense-core secretory granules, impairment of regulated secretion of a transfected prohormone, and reduction of secretory granul...
متن کاملVti1a functions in dense-core vesicle biogenesis
The SNARE-protein Vti1a is proposed to drive fusion of intracellular organelles, but recent data also implicated Vti1a in exocytosis. Here we show that vti1a is absent from mature secretory vesicles in adrenal CCs, but localizes to a compartment near the Trans-Golgi-Network, partially overlapping with Syntaxin6. Exocytosis is impaired in vti1a null cells, partly due to fewer Ca-channels at the ...
متن کاملSecretogranin II binds to secretogranin III and forms secretory granules with orexin, neuropeptide Y, and POMC.
Functional variations in the secretogranin III (SCG3) gene are associated with susceptibility to obesity. SCG3 forms secretory granules with orexin, melanin-concentrating hormone (MCH), neuropeptide Y (NPY), and POMC in the hypothalamus. In this study, we screened proteins for SCG3-binding activity and identified secretogranin II (SCG2) using a yeast two-hybrid system. Immunoprecipitation revea...
